An 44-year-old man has become the third man charged over the fatal shooting of former Melbourne underworld figure Mitat Rasimi in 2019.

The 51-year-old father-of-four was found unresponsive inside his car with multiple gunshot wounds after crashing into a pole in Dandenong in 2019.

Detectives arrested a Lysterfield South man this morning at his home just after 5am.

The man was interviewed by police and charged with one count of murder.

A 32-year-old Hallam man also arrested at his home was released pending further enquiries.

Detectives also executed a warrant at a gym in Hallam just after 7am as part of today's operation.

Items seized included a computer system.

It's understood the man charged today is connected to the gym.

Victoria Police Acting Deputy Commissioner Bob Hill said today's arrest was a direct result of the amount of work over the past few years by investigators.

"It's (been) one of the most significant arrests made by Victoria Police in many years," he said.

"This arrest is a testament to the hard work of police and investigators more broadly."

During the investigation, a 27-year-old man has since been charged with Rasimi's murder in November 2019.

Another man, 29, was also charged with murder in February 2020.

Police earlier said Rasimi had connections with a number of criminal organisations, including outlaw motorcycle gangs, but refused to give details.

Police have warned they will continue to pursue anyone involved in the attack.

"Some individuals and criminal groups think they can operate about the law – time and time again they are proven wrong," Assistant Commissioner Hill said.

"They may be able to avoid law enforcement for a period but it is only a matter of time before they face their day of judgement."

The 44-year-old man will appear at Melbourne Magistrates' Court via video link today.

Assistant Commissioner Hill said despite today's arrests, the investigation is continuing.
