A man has been shot dead on a busy inner-Melbourne street in what police believe is a targeted attack.

Police said the man approached an Audi, believed to have two people inside he knew, about 4.20pm on Tuesday in Fitzroy.

Detective Superintendent Paul O'Halloran said the man, believed to be 55, was shot as he left the dark-coloured sedan on Brunswick Street, after what police earlier described as an altercation.

Passersby found the man and called triple zero.

Ambulance crews treated the man, who is yet to be formally identified, but he died at the scene.

Police have cordoned off the area and a crime scene has been established

Police are still working to determine the exact circumstances of the incident, which they believe is not linked to recent gang-related shootings.

O'Halloran said it would appear the man had left housing commission flats in nearby Brunswick before approaching the car.

"At this stage, we don't know where the vehicle is," he said, adding that police didn't have the registration either.

O'Halloran said police could not confirm if the man lived in the commission flats.

He said the deceased man did not appear to have links to any outlaw motorcycle gangs or Middle Eastern organised crime groups.

Police are appealing for anyone with information or CCTV or dash cam footage to come forward.
